Nikon Spinning Disc TIRF STORM
Inverted spinning disc confocal microscope Inverted spinning disc confocal microscope for high speed optical sectioning. Spinning disc confocal microscopy offers several advantages over conventional optical microscopy, including wide-field and laser scanning confocal microscopy. This confocal technique allows acquisition of images at high frame rates. These qualities make Spinning Disc confocal microscopy particularly well suited to high speed 2D and 3D imaging of fixed and living cells with less photo-bleaching. A spinning disc confocal scanner uses rotating disks with thousands of pinholes typically arranged in an Archimedean spiral. Since the disc has a fixed pinhole size, this microscope is only truly confocal at 63 to 100x magnifications.
Super resolution using single molecule localization This microscope is also optimized for single molecule imaging. Stochastic Optical Reconstruction Microscopy (STORM) is one of the more common names for this super-resolution technique which provides some of the highest optical resolutions (12 nm in X an Y). STORM can be combined with another optical technique that this scope can do, Total Internal Reflection Fluorescence (TIRF) microscopy. TIRFM provides a second means of doing optical sectioning besides the spinning disc. This also allows one to do 3D STORM. TIRFM is typically limited to samples right up against the coverslip but HILO (Highly inclined and laminated optical sheet) TIRF allows one to go a few microns deep.
